We are leveraging our biologics and Chaperone-Advanced Replacement Therapy (CHART™) platform technologies to develop novel enzyme replacement therapy (ERT) products for Fabry disease, Pompe disease, and other Lysosomal Storage Disorders (LSDs). Our lead biologics program is ATB200/AT2221, a uniquely engineered Pompe disease ERT that will be administered in combination with a pharmacological chaperone.

Job Description:

Position Summary

This position will be responsible for the design and execution of purification / characterization of protein products expressed from mammalian and prokaryotic cell systems from bench scale to a multi-liter level. This resource will use a combination of conventional and proprietary ligand chromatography to achieve active, high-yield, high-purity biologics for evaluation in preclinical models. This resource will also share responsibility for the introduction, selection, propagation, and characterization of expression vectors in mammalian cell lines.

Roles and Responsibilities
Protein purification from prokaryotic and mammalian cell lines
Characterization of protein activity, purity, and yield
Creation, propagation, and characterization of mammalian cell lines expressing biologics
Documentation of all work in an electronic notebook
Analysis and presentation of results to larger scientific group

Qualifications: 

Requirements
BS or equivalent in a scientific discipline and a minimum of 2 years related lab experience, or MS or equivalent with 0-2 years’ experience
Proven track record for protein purification from prokaryotic and mammalian expression systems
Experience with protein characterization techniques including enzyme activity assays
Able to function both independently and as part of a team
Demonstration of detailed record keeping and data documentation

Experience and Skills
Prokaryotic and mammalian cell propagation, harvesting, and lysis.
Protein quantitation by multiple methods, SDS-PAGE, gel staining, and western blotting.Enzyme activity assays.
Purification scouting for standard and proprietary affinity resins.
Familiarity with Äkta purification systems preferred.Ability to summarize and evaluate protein purification strategies in terms of activity, purity, and yield.
