We are leveraging our biologics and Chaperone-Advanced Replacement Therapy (CHARTâ„¢) platform technologies to develop novel enzyme replacement therapy (ERT) products for Fabry disease, Pompe disease, and other Lysosomal Storage Disorders (LSDs). Our lead biologics program is ATB200/AT2221, a uniquely engineered Pompe disease ERT that will be administered in combination with a pharmacological chaperone.

Job Description:

Position Summary

The successful candidate will have broad experience with the full array skills employed in the biochemistry and molecular biology laboratory. The development, production, and characterization of biotherapeutic enzymes and proteins is an integral part of this position. Particular importance is placed on the skills to develop assays to evaluate different glycoprotein variants, enzymatic remodeling of glycoproteins, and process development for manufacturing of glycoproteins. Experience with non-antibody based therapies helpful.

Roles and Responsibilities
Develop lysosomal protein specific assays to evaluate N-linked oligosaccharides on glycoproteins (specialized phosphorylated oligosaccharides)
Develop specific reagents for the characterization of lysosomal glycoproteins
Perform analytical characterization during the purification process
Support assay transfer and process scale up
Develop and qualify analytical assays to support biological drug development
Serve on and/or lead departmental and inter-departmental teams
Write and review internal and external developmental reports

Qualifications: 

Requirements
Ph.D. in Biology or Biochemistry (or relevant discipline), and minimum of 5 years of glycobiology experience
Or
MS in Biology or Biochemistry (or relevant discipline), and minimum of 7 years of glycobiology experience
BS in Biology or Biochemistry (or relevant discipline), and minimum of 10 years of glycobiology experience
General biological lab experience
Expression of recombinant proteins using bacterial and mammalian expression systems
Enzymatic or chemical modification of glycoproteins
Structural N-linked oligosaccharide characterization of glycoproteins
Glycoprotein-receptor binding assay development
Enzymatic assay development – colorimetric, fluorescent, and isotopic
GXP experience
Experience with protein characterization including:
SDS-PAGE/Western-blot analysis
ELISA-based assays
Purification of proteins and enzymes using affinity, ion-exchange, and hydrophobic interaction chromatography using AKTA platforms
HPLC/UHPLC chromatography of peptides and small molecules and reverse phase chromatography
Steady-state and pre-steady-state kinetic analyses
